ACE-031 is a soluble form of activin receptor type IIB (ActRIIB) fused to the Fc region of human IgG1. Acts as a myostatin trap — binding and neutralizing myostatin and other TGF-beta family ligands (activins, GDF-11). Phase 2 data in Duchenne muscular dystrophy showed significant lean mass increases. 98.0% HPLC purity.

Mechanism of Action

Soluble ActRIIB-Fc decoy receptor sequesters myostatin, activin A/B, and GDF-11, preventing their binding to endogenous ActRIIB. This removes brake signaling on muscle protein synthesis, enabling rapid skeletal muscle hypertrophy in animal and early human studies.

Research FAQ

How does ACE-031 differ from myostatin antibodies?

ACE-031 neutralizes all ActRIIB ligands (myostatin, activins, GDF-11) simultaneously via a soluble receptor trap, unlike antibodies that target only myostatin. This broader ligand neutralization produces larger lean mass effects but also more systemic activity.
